mRNA Nanoparticle Therapy Shows Promise to Delay Type 1 Diabetes in Mice
University of Chicago researchers report in Cell Reports Medicine that they have developed an mRNA-loaded nanoparticle system that targets insulin-producing pancreatic beta cells and induces them to express PD‑L1, a protein that can shield cells from autoimmune attack. In mouse models of type 1 diabetes, the therapy reached the beta cells, boosted PD‑L1 production and delayed disease progression, and the approach also showed protective effects in mice transplanted with human beta cells. Lead author Jacob Enriquez, PhD, calls the work a proof of concept demonstrating both specific beta‑cell delivery and immune protection, while co‑author Mirmira says it opens the door to engineering beta cells rather than broadly suppressing the immune system. The team stresses that the study was done only in lab and animal models, that it did not examine long‑term safety or durability, and that substantial further testing will be needed before any human trials. If future human studies confirm safety and efficacy, the approach could offer a more targeted way to prevent or delay type 1 diabetes by protecting insulin-producing cells instead of globally modulating immunity.
